چکیده

We investigated whether pre- and posttreatment analysis of erythrocyte membrane Na+,K(+)-ATPase (EC 3.6.1.37) activity would be a useful marker for screening hypertensive patients to determine who might benefit from treatment with calcium antagonists. Erythrocyte Na+,K(+)-ATPase activity and sodium and potassium (ENa, EK) contents were determined in controls and in patients with untreated essential hypertension before and after 4 weeks of treatment with nitrendipine. Na+,K(+)-ATPase activity was significantly (P < 0.0001) less in untreated hypertensive patients (n = 15; 104.60 +/- 29.37 nmol of phosphate produced per milligram of protein per hour) than in controls (n = 15, 171.87 +/- 34.42). After 4 weeks of nitrendipine treatment Na+,K(+)-ATPase activity was greater than in the pretreatment group: 158.13 +/- 26.80 (P < 0.001). Pretreatment ENa contents (22.34 +/- 4.77 mmol/L) were significantly (P < 0.0001) higher than in the normotensive group (13.14 +/- 3.32 mmol/L), but there was no significant difference between the controls and the posttreatment group (14.84 +/- 3.49 mmol/L). The control and pretreatment groups showed negative correlations between enzyme activity and systolic/diastolic blood pressure (P < 0.0001). The control and the posttreatment groups showed an inverse correlation between enzyme activity and ENa contents: r = -0.608 (P < 0.05) and r = -0.724 (P < 0.001), respectively. Although Na+,K(+)-ATPase is restored in hypertensive patients receiving nitrendipine treatment, relative changes in enzyme activity in relation to relative reduction in blood pressure response to treatment were not correlated.

O-10: A Marked Animal-Vegetal Polarity in The Localization of Na+,K+-ATPase Activity and Its Down-Regulation Following Progesterone-Induced Maturation

Background: Polarized cells are key to the process of differentiation. Xenopus oocyte is a polarized cell that has complete blue-print to differentiate 3 germ layers following fertilization, as key determinant molecules (Proteins and RNAs) are asymmetrically localized. The objective of this work was to localize Na+, K+-ATPase activity along animal-vegetal axis of polarized Xenopus oocyte and fo...

Erythrocyte ghost Na+,K+-ATPase and blood pressure.

To examine the relationship between body mass index, blood pressure, and the Na+,K+-adenosine triphosphatase (ATPase) system, we measured the erythrocyte ghost Na+,K+-ATPase and the erythrocyte Na+ concentration in 120 blacks and 127 whites (136 males and 111 females). Blacks showed a 13.9% higher erythrocyte Na+ (7.63 +/- 0.19 vs 6.70 +/- 0.11 [SEM] mEq/L; p = 0.0001) and a 16.1% lower erythro...

inhibition convincingly to increased vascular smoothmuscle tone. Furthermore, any hypothesis which attempts to relate such a substance to the pathogenesis of essential hypertension must take into account, in addition to the problems outlined above, the fact that while there is consistent evidence of inhibition of Na+ ,K+-ATPase in the leucocytes of patients with essential hypertension, the body...

O-13: Na+/K+-ATPase Alpha1 Isoform Mediates Ouabain-Induced Expression of Cyclin D1 and Proliferation of Rat Sertoli Cells

Background: Novel roles for the interaction of cardiotonic steroids to Na+/K+-ATPase have been established in recent years. The aim of the present study was to investigate the intracellular signaling events downstream the action of ouabain on Na+/K+-ATPase in Sertoli cell obtained from immature rats. Treatment of Sertoli cells with ouabain (1 μM) induced a rapid and transient increase in the ex...

Race and sex differences in erythrocyte Na+, K+, and Na+-K+-adenosine triphosphatase.

Several reports indicate that erythrocytes (RBCs) from blacks and men have higher sodium concentrations than those from whites and women. One possible mechanism to explain this finding is a difference in the activity of Na+-K+-ATPase. To explore this possibility, we have studied the Na+ and K+ kinetics of RBC Na+-K+-ATPase and RBC Na+ and K+ concentrations in 37 normotensive blacks and whites, ...
